US 13 forms the commercial district around the State Capital, the City of Dover. Between Dover and Wilmington in New Castle County, US 13 is paralleled by the Delaware Route 1 Turnpike. US 13 crosses the Chesapeake and Delaware Canal on the St. Georges Bridge. US 13 continues toward Wilmington, sharing a concurrency with U.S. Route 40 in the New Castle area. US 13 bypasses the heart of Wilmington to the east, while U.S. Route 13 Business passes through the downtown area. Near its northern beginning, US Highway 13 Delaware parallels Interstate 495 between Wilmington and the Pennsylvania border.

In Delaware, U.S. Route 13 is known as the Dupont Highway from Wilmington to the Maryland state line. It composes the longest signed highway within the state of Delaware. This federal highway varies from four to six lanes as it careens through the Delaware cities/towns of Seaford, Laurel, Greenwood, Harrington, Camden, Dover, Smyrna, Odessa, Wilmington, and Claymont.

The origins of the Dupont Highway U.S. Route 13 predate the U.S. Highway system. T. Coleman du Pont, an engineer of the famous du Pont family, envisioned a paved north-south statewide roadway. Coleman proposed the project to the citizens of Delaware in 1908. His concept entailed the creation of separate north and southbound lanes, trolley track lines, and a pathway for horse-drawn vehicles. In 1924 a two-lane version of his visionary highway project opened to traffic between the southern agricultural areas and the urbanized center of Wilmington. The road was a huge success and in 1933 the Delaware State Highway Department expanded the highway.

US 13 was the world's first divided highway.
